SUMMARY:

The increasing use of plant-based ingredients in the diets of aquatic organisms reinforces the importance of phytase in increasing phytate-phosphorus bioavailability, reducing inorganic phosphorus supplementation, and minimizing environmental impacts. This dissertation was structured into two studies: a scientometric survey on the use of phytases in diets for fish and shrimp, and the production and characterization of a novel fungal phytase from isolates of the genus Mucor. In the first study, a scientometric analysis was performed on literature indexed in the Web of Science database, using the descriptors phytase AND fish and phytase AND shrimp, covering the period from 2014 to 2024. A total of 187 documents were analyzed using bibliometric techniques with the aid of VOSviewer and Bibliometrix software. The results showed growth in scientific production on the use of phytases in aquaculture, with Pakistan standing out as the main knowledge producer and the journals Aquaculture and Aquaculture Nutrition as the main publication outlets. In addition, the main scientific collaboration networks, keywords, and research trends were identified, revealing the low representativeness of studies focused on shrimp farming. In the second study, six fungal isolates from the semi-arid region of Pernambuco were evaluated for phytase production via solid-state fermentation using wheat bran as substrate. Mucor lusitanicus URM7853 showed the highest phytase activity (53.74 U/mL) and was selected for biochemical characterization. The enzyme showed optimal activity at pH 5.0 and a temperature of 40 °C. Thermal stability was assessed between 50 and 80 °C, with the longest half-life observed at 50 °C (990.21 minutes), indicating potential for application in animal feed drying. The kinetic parameters determined were a Km of 24.16 mM and a Vmax of 83.33 mM/min, along with increased enzymatic activity in the presence of Ca²⁺ and Fe²⁺ ions. It is concluded that the scientometric survey demonstrates the expanding scientific interest in the use of phytases in aquaculture and the need for further studies, especially in shrimp farming. Additionally, the phytase produced by M. lusitanicus URM7853 showed promising biochemical characteristics, indicating potential for future applications in aquatic organism nutrition and contributing to the development of more sustainable strategies for aquaculture.
