Donkey milk has attracted scientific and commercial interest due to its nutritional profile and therapeutic potential. Its composition, similar to that of human milk, makes it a promising alternative for infant nutrition and for individuals with cow’s milk protein allergy. This study aimed to evaluate the effect of dietary supplementation with different oils on the chemical composition of milk from donkeys of the Northeastern ecotype, with emphasis on the modulation of the fatty acid profile. Sixteen lactating females of the Northeastern ecotype were used in a completely randomized experimental design, consisting of four treatments: a control diet without oil inclusion and three diets in which 20% of the energy provided by the grains in the control diet was replaced with soybean oil, canola oil, and marine algae oil. After a 25-day adaptation period to the experimental diets, milk samples were collected over five consecutive days for chemical composition analysis. Data were subjected to analysis of variance, and means were compared using Tukey’s test (P < 0.05). No differences (P > 0.05) were observed between the control group and the treatments with different lipid sources for total solids, protein, fat, lactose, and ash, indicating stability in milk nutritional composition. Regarding the fatty acid profile, modulation of the lipid fraction was observed, with an increase in the proportion of unsaturated fatty acids in the oil-supplemented treatments. Notably, oleic acid (C18:1) increased in the canola oil treatment, while linoleic acid (C18:2) increased in the soybean oil and marine algae oil treatments, resulting in a higher unsaturated-to-saturated fatty acid ratio. It is concluded that the inclusion of different lipid sources in the diet of donkeys improves the milk fatty acid profile, making it more similar to human milk. Among the evaluated sources, soybean oil stands out for its better cost-effectiveness and its potential to produce milk with a more favorable lipid profile.
