The objective of this study was to determine the total production and composition of milk in female donkeys of the Nordestino ecotype. 14 female donkeys and their foals were used, in a completely randomized experimental design, in a split-plot scheme, with the plots consisting of two groups (primiparous females and multiparous females) and the subplots consisting of the evaluation periods. The total milk production of females was determined by weighing the foals, before and after feedings, at 20, 40, 60, 80, 100, 120, 140, 160 and 180 days after birth. The physical-chemical composition of the milk was determined on days 1, 3, 15, 30, 60, 90, 120, 150 and 180 after birth. The measurements of the foals were carried out by taking 15 linear measurements. The results were subjected to analysis of variance, mean tests, regressions, correlations and multivariate principal components. There was no difference between the groups of primiparous and multiparous women for total milk production, daily evaluation times, milk pH and lactose and milk protein levels. The donkeys produced an average of 5.00 kg of milk, with peak production at 60 days of lactation. The donkey milk samples presented a pH close to neutrality and, throughout lactation, the protein content of the donkey milk samples progressively reduced, going from 4.44 g/100 mL on the first day of lactation to 1.55 g/100 mL. mL at 180 days after birth. Regarding the lactose content of milk, lower concentrations were observed in colostrum and transitional milk and higher values between 15 and 120 days of lactation, with a maximum recorded value of 7.77 g/100 mL. Regarding the fat content of milk, on the first day of lactation an average fat content in milk of 1.65 g/100 mL was observed and between 3 and 120 days of lactation it ranged from 0.45 to 0.73 g. /100ml. The relationship between milk production and linear measurement gain of foals showed progressive growth. Furthermore, there was a correlation between milk protein and the height at the withers, height of the rump and chest perimeter of the foals, with values of 65.39; 64.03; 64.57 and 61.16, respectively. It is concluded that the donkey of the Nordestino ecotype has potential for dairy production, and the nutritional characteristics of the milk favor its use as a nutraceutical food.
