Donkey milk has gained commercial interest due to its nutritional profile and therapeutic potential. Its composition, similar to that of human milk, makes it a promising alternative for infant nutrition and groups with special needs. The objective was to evaluate the effects of including different lipid sources in the diet of donkeys of the Northeastern ecotype on the fatty acid profile and nutritional composition of their milk. Sixteen female donkeys of the Northeastern ecotype were used, distributed in a completely randomized experimental design, consisting of four treatments: a control diet without added oil and three diets supplemented with soybean oil, canola oil, and algae oil. After 25 days of adaptation to the experimental diets, milk samples were collected for five consecutive days to evaluate the physicochemical composition and determine the fatty acid and protein profiles. The data were subjected to analysis of variance, and the means were compared using Tukey's test (P < 0.05). There was no difference (P > 0.05) between the control group and the treatments with different lipid sources in the levels of total solids, protein, fat, lactose, and ash, indicating stability in the nutritional composition of the milk in addition to the low somatic cell count. Regarding the fatty acid profile, modulation of the lipid fraction was observed with an increase in the proportion of unsaturated fatty acids in the treatments supplemented with oils, with emphasis on oleic acid (C18:1) in the treatment with canola oil and linoleic acid (C18:2) in the treatments with soybean oil and seaweed oil, resulting in an increase in the unsaturated:saturated ratio. It is concluded that the inclusion of different lipid sources promotes improvement in the lipid profile of milk, with an increase in the fraction of unsaturated fatty acids.
